Title:Effects of Olanzapine in Reducing the Emergence of Psychosis Among Nursing Home Patients With Alzheimer's Disease.

Abstract: This article examines the effects of olanzapine on the emergence of psychotic symptoms among nursing home residents with Alzheimer's disease (AD). A total of 206 residents were enrolled in a multicenter, placebo-controlled, double-blind study. Participants received either placebo or fixed-dose olanzapine (5, 10, or 15 mg/day) for up to 6 weeks. This analysis uses data for a subset of patients without clinically significant hallucinations (n=153), delusions (n=87), or either symptom (n=75) at baseline. Of the patients without hallucinations or delusions at baseline, significantly more psychotic symptoms emerged among those treated with placebo than those treated with olanzapine. Similarly, among patients without baseline hallucinations, those treated with placebo experienced more hallucinations than those treated with olanzapine. Patients without delusions at baseline showed no significant treatment effects. Olanzapine had a favorable safety profile in all patient subsets. The results suggest that olanzapine may attenuate the emergence of psychotic symptoms in patients with AD. 1 figure, 4 tables, 40 references. (AA-M).
